A sandwich-type ELISA was developed for assessment of a complex between cathepsin B (CB) and cystatin C (CC) in sera. The assay is based on a combination of two monoclonal antibodies, first one (3E1) - catching Ab, was raised against CB and the second one (1A2)-detecting Ab, conjugated with horseradish peroxidase, against CC. Only CBžCC complex, cross-linked and non cross-linked, was able to evoke the signal in ELISA. Detection limit of the assay was 31.25 nmolž1 and the working range between 31.3 - 400 nmolž1. The within and between-run coefficient of variance (CV) varied from 4.7% to 9.4% and from 11 % to 12.8%, respectively, indicating satisfactory reproducibility of the method. The content of CBžCC complex was determined in sera of 133 healthy controls, 32 patients with benign lung diseases, 148 with lung and 32 with colorectal cancer.
