S spoynanjem, da so dolořeni genotipi HPV tesno poveyani y nastankom raka materniřnega vratu, se je porodila ideja, da bi y odkrivanjem okuÍb s HPV lahko prepoynali Íenske, ki so v nevarnosti ya rayvoj predrakastih in rakastihsprememb na materniřnem vratu. Opisanih je bilo veř metod ya odkrivanje okuÍb s HPV, vendar nobena ni iypolnila vseh priřakovanj. V naǴi rayiskavi smo primerjali uřinkovitost komercialno dostopne prve generacije Digene Hzbrid Capture testa I (HC I), ki yaynava 14 mejno- oyiroma visokoriyiřnih genotipov HPV (HPV-6, -11, -16, -18, -31, -33, -35, -42, -43, -44, -45, -51, -52 in -56) in njene iyboljǴane rayliřice, Digene Hzbrid Capture testa II (HC II), ki yaynava Ǵe Ǵtiri dodatne genotipe (HPV-39, -58, -59 in -68). V rayiskavo smo vkljuřili 483 nakljuřnih yaporednih vyorcev brisov materniřnega vratu, ki so bili odvyeti enakemu Ǵtevilu Íensk y abnormalnimi iyvidi citoloǴkih presejalnih testov po Papanicolaou. Vse vyorce smo vyporedno testirali y obema testoma na prisotnost mejno- in visokoriyiřnihgenotipov HPV. Reyultati testov so se ujemali v 468 (96.8%) od 483 vyorcev; 193 vyorcev je bilo HPV DNA negativnih in 275 HPV DNA poyitivnih.Pri 15 vyorcih se reyultati obeh testov niso skladali; vyorci so bili poyitivni le pri testiranju s testom HC II. Da bi dolořili status DNA HPVv teh vyorcih, smo jih dodatno testirali s skupinsko ynařilno MZ09/MZ11 L1 HPV veriÍno reakcijo s polimerayo (angl. polzmerase chain reaction, PCR). 14 od 15 vyorcev je bilo jasno PCR poyitivnih. Genotipiyacija y encimsko raygradnjo pridelka PCR s sedmimi restrikcijskimi endonukleayami je pokayala, da so bili v 7 od 14 vyorcev prisotni HPV genotipa 16, v 2 HPV genotipa 56, v ostalih pa HPV genotipa 18, 31, 35, 58 oyiroma 68. Glede na reyultate opravljene Ǵtudije je Digene Hzbrid Capture test II obřutljivejǴi od njegovegapredhodnika, Digene Hzbrid Capture testa I. Poveřano obřutljivost HC II v veřji meri pripisujemo spremembi sestave reagentov in v manjǴi meri dodatku novih RNA lovk ya Ǵtiri dodatne genotipe HPV. Digene Hzbrid Capture test II je tudi hitrejǴi in laÍje iyvedljiv in yato bolj primeren ya rutinsko dokayovanje okuÍb s HPV.

The human papillomaviruses (HPV) are small DNA viruses that belong to the family Papovaviridae. Infection with certain genotypes of HPVs are intimately linked with squamous epithelial cell benign and malignant lesions of the lowergenital tract in both men and women. The role of HPV infection in etiology of oral squamous cell carcinoma still remains inconclusive, since theprevalence rates of HPV infection obtained in different studies varied from0% to 76.4%. In our study we investigated the possible role of HPV infection in the etiology of oral squamous cell carcinomas obtained from Slovenian patients. Using three different polymerase chain reaction (PCR) protocols and in situ hybridization (ISH) the presence of HPV DNA was comparatively investigated in 41 archival tissue specimens of oral squamous cell carcinoma obtained from the same number of patients (representative population of Slovenian oral squamous cell carcinoma patients) and in 41 histologically normal oral mucosa specimens obtained from the same number of subjects, who matched the case subjects in age, sex, localization of obtained tissue specimens, drinking and smoking habits. According to the available literature data, our investigation is the second study in which HPV-DNA prevalence was comparatively studied in oral squamous cell carcinoma patients and matching control subjects and the first study where the presence of HPV-DNA was comparatively studied on tissue samples in both groups. Using all three PCR protocols HPV-DNA was detected in 3 out of 41 (7,3%) oral squamous cell carcinoma specimens. Tissue specimens contained HPV-16, HPV-33 and HPV-58. Using ISH only one HPV-DNA positive specimen was identified. To the best of our knowledge, our HPV-33 and HPV-58 positive oral squamous cell carcinomas represent the first oral carcinomas in which these two HPV genotypes were detected.
